Expressing Recombinant Genes In Vivo

چکیده

genes in specific cell types in vivo would help to determine their mechanism of action. We describe a method to transfer vascular smooth muscle cells expressing recombinant gene products to localized segments of the arterial wall. Vascular smooth muscle cells from the inbred Yucatan minipig were infected in vitro with an amphotropic, replication-defective retrovirus transducing the gene for Escherichia coli P-galactosidase. Vascular smooth muscle cells expressing this recombinant gene were implanted, using a catheter, into denuded iliofemoral artery segments of pigs in vivo. These arteries subsequently demonstrated J-galactosidase activity in cells of the intima and media. This method, which provides for the introduction of genetically modified smooth muscle cells, can be used to define the biological effects of recombinant genes in the vessel wall and potentially to provide alternative treatments of vascular diseases. Construction of the Recombinant Plasmid Expressing AID under the Control of Temperature-sensitive Promoter of Bacteriophage Lambda

Background and purpose: Activation-induced cytidine deaminase (AID) is a B-cell specific enzyme responsible for somatic hypermutation (SHM) and class switch recombination (CSR) of antibody genes within the B-cell follicle of peripheral lymphoid organs. Ectopic overexpression of the enzyme leads to mutations in non-B cells and Escherichia coli (E.coli) genes. However, induction of mutations in E...

Expressional Comparison Between Episomal and Stable Transfection of a Selected tri-fusion Protein in Leishmania tarentolae

Introduction: Leishmania tarentolae (L. tarentolae) is a nonpathogenic species of Leishmania genus that can be used as an expression system to produce immunogenic proteins or epitopes in vivo acting as an efficient and safe recombinant live vector in vaccinology. Cysteine proteases (CPs) are one group of Family C1 peptidases in Leishmania that are important for survival of the parasite within...

Construction of recombinant yeast expressing EpEX as a suitable candidate in cancer diagnosis and therapy

Introduction: Epithelial cell adhesion molecule (EpCAM) is a membrane glycoprotein that is overexpressed on the majority of tumor cells of epithelial origin and thereby can be used as a target of immunodetection and immunotherapy of cancer. So, it is important to produce this protein in its native form. Interestingly, during the last two decades, the yeast Pichia pastoris (P. pastoris) has beco...

Evaluation of the Effect of Promoter Type on the Immunogenicity of the Live Recombinant Salmonella Vaccines Expressing Escherichia Coli Heat-labile Enterotoxins (LTB)

Enterotoxigenic Escherichia coli (ETEC)-induced diarrhoea is the second most commoncause of death in children in the developing countries. Heat labile toxin (LT) is responsible forETEC-induced diarrhoea. In the present study, a novel live ETEC vaccine based on subunitB of LT (LTB) expression in attenuated PhoPc Salmonella strain was developed. Herein, weaimed to compare the in-vitro activity of...
